Hmmm.....I've tried this on Windows XP Pro and you can cartainly change the 
USB drive letter but it may more may not remain the same when you remove it 
and put it in again.  If you use a different USB drive in that same slot 
(even the same brand) the drive letter is subject to change.

This has been my experience.  So, probably some locking utility might be the 
answer.
